Genome-wide Association Study of Genetic Loci and Alzheimer Disease

To the Editor: The multistage genome-wide association study (GWAS) analysis of genetic loci associated with Alzheimer disease (AD) conducted by Dr Seshadri and colleagues1 presented evidence for association of 2 novel genetic loci with AD as well as confirmation for 2 previously reported associations (CLU and PICALM). rs744373 is located on chromosome 2, in a region that to our knowledge has not previously been associated with late-onset Alzheimer disease; there are no linkage entries for Chr 2 in http://www.alzgene.org or http://alzres.com.

The one published exception is a study2 that stratified the National Institute of Mental Health Alzheimer Disease Genetics Initiative data set by autopsy status. Linkage analysis limited to autopsy-only sibling pairs identified several unique linkage regions throughout the genome, including one on chromosome 2, centered at 110 cM (half-height range, 90-140 cM), that encompasses rs744373.
