Transcranial focused ultrasound (tFUS) is a promising technique for non-invasive brain stimulation. By transmitting low-intensity ultrasound waves, tFUS can target deep brain regions with potentially greater precision than other forms of non-invasive brain stimulation.
As the brain is involved in pain processing and perception, researchers want to learn whether targeting specific brain structures with tFUS stimulation can potentially change how the brain processes pain.
In a preclinical study published in Blood, a collaborative research team designed a new multi-element ultrasound device to target specific brain regions in mice that process pain. The researchers used a genetically engineered mouse model of sickle-cell disease, a painful human condition characterized by sickle-cell shaped red blood cells. This condition leads to heightened sensitivity to different types of pain and was chosen to determine if this ultrasound approach could impact sensitivity to pain.
Treatment with the new device showed a significant decrease in pain hypersensitivity, particularly in response to heat-induced pain in the mouse model.

Unlicensed dispensaries and smoke shops are significantly less likely than licensed retailers to require age verification before somebody enters a store or makes a purchase, according to a secret shopper study published today in Pediatrics. Shoppers went to 37 dispensaries and smoke shops in New York City, also finding that unlicensed stores more often sold products that appeal to young people like energy drinks, soda, and candy.
Part of the point of legalizing weed is to reduce risk through regulation, the authors write. But in New York, unlicensed retailers outnumber the licensed, regulated ones. In a First Opinion essay last year, three researchers and advocates wrote that, “implementing strong and comprehensive protections for children is not the norm in many states that have legalized recreational marijuana use.”

The factors influencing when people are diagnosed with dementia
People with lower levels of education, who live alone or in a care home, or who don’t have a close relative, are about three years older when they receive an official dementia diagnosis compared to their counterparts, according to a study published Friday in JAMA Network Open that looked at more than 107,000 people in Sweden who were diagnosed with dementia between 2018 and 2022. On top of that, people who had less than a high school education (fewer than nine years) had more comorbidities and took more medications than those with more schooling.
The results could indicate that these populations tend to have more advanced disease or a poorer prognosis, the authors write. They couldn’t say what caused the delayed diagnoses, of course, but hypothesized that the risk of losing a driver’s license may play a larger role in when to seek care for people who live alone or have fewer family members.

Can treatments for lung conditions go deeper?
Between common chronic conditions like COPD and asthma, and acute harm inflicted by viruses, lung damage is a big problem. Many treatments focus on the upper airways. But to go deeper, researchers are studying the pair found in some Covid vaccines: mRNA + a lipid nanoparticle.
New research in mice, published Friday in Nature Communications, posits that a similar technology could be used to stimulate healing in parts of the lung not easily reached by conventional therapies. The team reports a nanoparticle, called ionizable amphiphilic Janus dendrimer, targets mRNA therapy directly to the lung and promotes tissue repair.
“If introduced early in the inflammatory process,” this mechanism could speed up healing, “ultimately reducing the incidence of complex lung injury and disease,” write the authors (several of whom are named on patents and patent applications related to the research). — Isabella Cueto

Portable MRI, enhanced by AI, proves viable in brain imaging for dementia: The low image quality of small, affordable MRI machines have prevented their widespread use. But a boost from AI could close the gap, bringing MRI to more patients.
Of all the instruments in the modern medicine toolbox, magnetic resonance imaging, or MRI, is one of the most powerful. But in many regions, the expense, space, and expertise required to use it puts this tool out of reach. A new study suggests that, with a performance boost from artificial intelligence (AI), existing portable, low-cost MRI tech could make high-quality brain imaging available to more patients.
The new system employs AI techniques to improve the quality of, and then automatically analyze, images taken from low-field strength MRI devices that, due to being smaller, sacrifice image quality compared to conventional MRI machines. In a recent clinical study, researchers from Massachusetts General Hospital, Harvard Medical School, and Yale School of Medicine used their automated approach to detect Alzheimer’s disease markers in humans, finding that their technique performed at the same level as higher-power MRI. Their results, published in Nature Communications, point to the new method as a potential means of providing broad access to high-quality brain imaging.

Among people aged 65 and older, half visited the emergency department (ED) in the last month of their lives, and three in four visited the ED during the 6 months before their deaths. However, emergency care has not adapted to meet these patients’ needs, with palliative care teams, present in two-thirds of hospitals, not typically available in EDs for crisis response.
